Mai was born as the older sister of Rin Nohara, whom she deeply loved and came to be a best friend of sorts to, as well as Saya Kirintei. She grew up to become a good friend of Team Minato, often spending time with them and helping Rin and Saya train. She was shown breaking up fights between Obito and Kakashi often.
Around the time of the battle of Kannabi Bridge, the mission during which Obito was lost on, Mai was sent on a mission, having been promoted to ANBU rank around the time she was sixteen. When she returned, she received news of Obito's death.
Some time after, Kakashi and Rin were sent on a mission. During this time, Mai remained in Konohagakure in order to provide back-up, if necessary. When she received no reports from anyone and no calls for back-up, she headed out to find what was wrong. It was then that she happened upon Kakashi, finding his way back to the village without Rin. When she asked him what had happened, he didn't respond and Mai understood what had happened to her sister. She returned to the village with Kakashi and according to Kakashi, never spoke to Kakashi again.
Mai is a bubbly, talkative type of girl. Although she is often shy around people she doesn't know, she quickly becomes comfortable around them as she knows them longer. She tends to be described as being eccentric, since she does weird things, according to Saya.
Mai looks like an older version of Rin and seems to represent what Rin would have looked like, had she been able to grow into an adult. Nevertheless, Mai has some different features than Rin, such as her blue eyes and longer brown hair.
Mai possesses the markings of the Nohara Clan, and is rarely seen without them, though as a chūnin she was seen without them from time to time. She typically wore a dark black zip-up tunic, dark black pants similar to those once worn by Hinata Hyūga, and traditional shinobi sandals. She typically doesn't wear her forehead protector. She also tended to wear earrings similar to those worn by Team Asuma.
